The aim of the present study was to investigate the effect of obesity and aerobic exercise, as well as the use of curcumin supplementation on inflammatory factors in male rats.
25 adult male Wistar rats, eight weeks old and weighing 187.5 ± 9.37 g, were randomly selected and to induce obesity, the rats were placed on a high-fat diet for 8 weeks. The rats were divided into five groups: healthy, obese, obese + exercise, obese + supplement, and obese + exercise + supplement. Curcumin supplementation and aerobic exercise were applied to the relevant groups for 8 weeks, 5 sessions per week. Real Time-PCR was used to measure the expression of TLR4, NF-κB, NLRP3, and IL-1β genes. One-way analysis of variance and Tukey's post hoc test were used to determine the difference between groups at a significance level of p < 0.05.
According to the results of the one-way analysis of variance test, there is a significant difference between the gene expression levels between the groups in the present study after eight weeks of aerobic exercise and curcumin supplementation (p=0.001). The results of the Tukey post hoc test showed that the gene expression levels in the obese group were significantly higher than in the healthy group (p=0.001). These gene expression levels were significantly lower in the obese+exercise, obese+supplement, and obese+exercise+supplement groups than in the obese group (p=0.001).
The present study suggests that aerobic exercise combined with curcumin supplementation leads to an improvement in the TLR4/NF-κB/NLRP3/IL-1β signaling pathway in obese male rats.
