Background : High-intensity circuit resistance exercise (CRE) can increase oxidative stress and inflammation, influencing apoptotic pathways in neutrophils. In contrast, supplementation with medicinal plants may accelerate neutrophil cell death by modulating these pathways. Given the role of Bcl-2 family proteins in regulating cell death, this study aimed to investigate the effect of acute jujube (Ziziphus jujuba) supplementation before an intense circuit resistance training session on the protein levels of Bcl-2, Mcl-1, Bid, and Caspase-9 in human neutrophils.
Materials and Methods: In this quasi-experimental study, 14 healthy young men were randomly assigned to two groups: exercise with placebo and exercise with Ziziphus jujuba solution (0.5 g per kg of body weight). Blood samples were collected at three time points, including pre-test (before supplementation), immediately after exercise, and 120 minutes post-exercise. Neutrophils were isolated using procedures, and the levels of Bcl-2, Mcl-1, Bid, and Caspase-9 were measured using human ELISA kits. Data were analyzed using two-way repeated-measures ANOVA (p≤0.05).
Results: The results showed that the time × group interaction effect was not significant for Bcl-2 and Mcl-1. For Bid, a moderate but non-significant interaction effect was observed. In contrast, a significant time × group interaction effect was found for Caspase-9 (p≤0.05), indicating that Ziziphus jujuba supplementation affected the protein level of Caspase-9 over time.
Conclusion: It appears that Ziziphus jujuba may contribute to the modulation of neutrophil apoptotic responses and support immune adaptation following intense exercise. However, studies with larger sample sizes and longer supplementation periods are necessary to confirm these findings.
